Some background: antioxidant enzymes areregulated at the DNA/genetic level by the antioxidant responsiveelement (ARE). Nrf2 (pronounced “nerf-2”) is an importantprotein thatremains dormant within each cell of our body, unable tomove orperform its task, until released by an Nrf2 activator. When Nrf2isreleased by an activator it migrates to the cell nucleus and bonds with the DNA at the place where the ARE resides. In this way Nrf2 activation provides a pathway for the production of our body’s mostessential antioxidants. This hypothesis hasbeen supported in many studies.

Nrf2is normally bound in the cytoplasm of cells to a protein calledKEAP1.However, when an appropriate phytochemical agent attaches to a kinase receptor on the cell wall a phosphate group is released thatcauses theNrf2 to be released. The Nrf2 then migrates into the cellnucleus andcauses an antioxidant enzyme to befabricated andreleased. This endogenously produced enzyme then canprotect against ROS, electrophiles, and other toxic agents. Inpractical experience, it has been found that a combination of multiplepolyphenols workssignificantly better than single ingredients. Oxygen radical absorbance capacity (ORAC) has been the traditionalmethod of measuring antioxidant capacities in biological samples.
